There is no lock. Just remove your rig, be it a single fly or fly, shot, strike indicator, and put it back in the rod case. Never put a fly rod back in the case with the fly in the hook keeper on the rod, it WILL get snagged in the fabric in the rod tube and then you have a nice sharp hook scratching your fly rod every time you put it in or remove it from the rod case.
If you don’t have a rod case, let me just say that the vast majority of rod breakages occur during transport to or from your fishing destination. A rod case costs $30 and is worth every penny.

I’ve never seen one lock. I don’t think there is a way. I assume by lock you mean have the spool not turn. You can crank the drag all the way down but it’s not necessary. Just get a reel case. Have your rod in a tube and your reel in a case and you are keeping them from getting banged up while waiting to use them again.

You do not want it to lock on flying rod because you want the luke to be moving at all the time for Bass or Trout to think it is alife not a dead bug fload on top of water if we have locked it all of our jugs, baits what ever is on the line will stay still and fish are so smart they know it is fake so have to let and give it some room to move not alot.. just have to pratice on it… Soon or later you will see Hey we got nice bass or trout fish..

You cant "lock" a fly reel. Just remove your leader as it can cut into your fly line and take the drag fully off to stop wear on the disc drag system, depending on your reel its usually done by twisting it anti-clockwise. There’s no need to remove the spool, you know when the spool is properly attached as you should hear it click on.
